Chloride Profile Analysis

Chloride profile analysis is the measurement and interpretation of chloride concentration at varying depths within a material, most commonly concrete, to assess chemical ingress and durability. In reinforced concrete, chloride ions migrate through pores by diffusion, convection, and capillary absorption, producing a concentration gradient that can be quantified from depth-based samples and modeled using transport equations such as Fick’s second law. Engineers use these profiles to estimate chloride penetration, evaluate the risk of reinforcement corrosion, and compare concrete mixtures, protective treatments, or exposure conditions. The analysis supports service-life prediction, maintenance planning, and the design of structures for marine, deicing-salt, and other chloride-rich environments.

Eukaryotic Polyribosome Profile Analysis

This article describes a protocol for the extraction of translating ribosomes from eukaryotic cells. Once extracted, ribosomes are separated into monosomes and polyribosomes by sucrose gradient fractionation to allow different ribosomal populations to be analyzed. As such, this method is the gold standard for examining the regulation of translation.

Metabolic Profile Analysis of Zebrafish Embryos

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

Quantification of Hydrogen Concentrations in Surface and Interface Layers and Bulk Materials through Depth Profiling with Nuclear Reaction Analysis

We illustrate the application of 1H(15N,αγ)12C resonant nuclear reaction analysis (NRA) to quantitatively evaluate the density of hydrogen atoms on the surface, in the volume, and at an interfacial layer of solid materials. The near-surface hydrogen depth profiling of a Pd(110) single crystal and of SiO2/Si(100) stacks is described.

Analysis of Translation Initiation During Stress Conditions by Polysome Profiling

Here, we describe a method to analyze changes in the initiation of mRNA translation of eukaryotic cells in response to stress conditions. This method is based on the velocity separation on sucrose gradients of translating ribosomes from non-translating ribosomes.
