Diaphragm Valve

A diaphragm valve is a fluid-control device that regulates or isolates flow using a flexible diaphragm, making it valuable when reliable sealing and contamination control are important. When an actuator or handwheel moves the diaphragm against a valve seat or weir, the membrane changes the passage area while separating the operating mechanism from the process fluid. This design supports tight shutoff and reduces leakage and product contamination, particularly in chemical processing, pharmaceutical manufacturing, water treatment, and sanitary systems. Diaphragm valves can also handle corrosive, viscous, or particulate-containing fluids, although their pressure and temperature limits depend on the diaphragm and body materials.

Diagnostic Ultrasound Imaging of Mouse Diaphragm Function

Diagnostic ultrasound imaging has proven to be effective in diagnosing various respiratory diseases in human and animal subjects. We demonstrate a comprehensive ultrasound protocol utilized by Dr. Zuo's lab to analyze diaphragm kinetics specifically in mouse models. This is also a non-invasive research technique which can provide quantitative information on mouse respiratory muscle function.

Functional and Morphological Assessment of Diaphragm Innervation by Phrenic Motor Neurons

Compound muscle action potential recording quantitatively assesses functional diaphragm innervation by phrenic motor neurons. Whole-mount diaphragm immunohistochemistry assesses morphological innervation at individual neuromuscular junctions. The goal of this protocol is to demonstrate how these two powerful methodologies can be used in various rodent models of spinal cord disease.

This protocol describes the isolation, culture, and calcification of rat-derived valve interstitial cells, a highly physiological in vitro model of calcific aortic valve disease (CAVD). Exploitation of this rat model facilitates CAVD research in exploring the cell and molecular mechanisms that underlie this complex pathological process.
