Low-input Library

Low-input library preparation is a sequencing method for converting very small amounts of DNA or RNA into a molecular library suitable for genetic analysis. The process typically fragments or converts the starting material, attaches platform-specific adapters, and uses limited-cycle amplification to enrich molecules while preserving sequence information and minimizing material loss. This approach enables next-generation sequencing when samples are scarce, degraded, or difficult to obtain, including single cells, rare clinical specimens, and precious biological materials. Low-input libraries support variant detection, transcriptome profiling, and studies of genetic diversity while helping researchers make efficient use of limited samples.

Contamination during the genomic sequencing of microscopic organisms remains a large problem. Here, we show a method to sequence the genome of a tardigrade from a single specimen with as little as 50 pg of genomic DNA without whole genome amplification to minimize the risk of contamination.

Methods for generating large-scale gRNA libraries should be simple, efficient and cost-effective. We describe a protocol for the production of gRNA libraries based on enzymatic digestion of target DNA. This method, CORALINA (comprehensive gRNA library generation through controlled nuclease activity) presents an alternative to costly custom oligonucleotide synthesis.
