Nascent Cdna Analysis

Nascent cDNA analysis is a molecular approach for examining newly synthesized complementary DNA (cDNA) to characterize the RNA transcripts produced by genes. In this process, reverse transcriptase copies RNA into cDNA, which can then be amplified, sequenced, or quantified to assess transcript identity, abundance, and processing. In genetics, analyzing nascent cDNA helps distinguish active gene transcription from changes in RNA stability and can reveal alternative splicing, transcriptional regulation, and emerging transcript variants. These measurements support studies of gene expression, developmental regulation, disease-associated mutations, and cellular responses to environmental or experimental conditions.

The cDNA microarray PtGen2 was developed for gene expression studies in loblolly pine, P. taeda, and other conifer species. Here, we show pre- and post-hybridization handling and washing techniques that can be used with this array to yield better consistency, reduced artifacts, and lower backgrounds.
