Fusion Transcript Detection

Fusion transcript detection is the identification of RNA molecules formed when segments from two distinct genes become joined, providing evidence of gene rearrangement, abnormal transcription, or pathogen–host interactions. The process typically converts RNA into complementary DNA and then uses targeted amplification, sequencing, or alignment methods to recognize junction-spanning sequences that distinguish fusion transcripts from normal gene products. In immunology and infection research, these analyses can reveal oncogenic rearrangements in immune cells, characterize microbial or viral integration events, and clarify how genetic alterations influence cellular signaling. Accurate detection supports disease classification, biomarker development, and investigation of mechanisms underlying infection-related and immune-mediated disorders.

A Cre-Lox P Recombination Approach for the Detection of Cell Fusion In Vivo

A method to track cell fusion in living organisms over time is described. The approach utilizes Cre-LoxP recombination to induce luciferase expression upon cell fusion. The luminescent signal generated can be detected in living organisms using biophotonic imaging systems with a sensitivity of detection of ˜1,000 cells in peripheral tissues.

Detection of circulating ribonucleic acid (cRNA) from blood is an unmet need in clinical diagnostics. Here we describe methods that characterize cRNA from non-small cell lung cancer patients using sensitive and specific digital polymerase chain reaction. The tests meet design requirements to detect fusion variants within 72 hours.

In this video, we demonstrate the electrophoretic mobility shift assay to detect protein-DNA interactions using infrared fluorescent-labeled DNA probes. When the protein binds to the DNA probe, it forms a large complex that migrates slowly on and appears as a shifted band on the gel.
