Luciferase-transduced A20

Luciferase-transduced A20 cells are A20 lymphoma cells engineered to express luciferase, creating a traceable model for studying tumor behavior and immune responses in vivo. After administration of a luciferase substrate, the enzyme catalyzes a light-producing reaction, and emitted bioluminescence can be detected to estimate cell presence, distribution, and changes in tumor burden over time. This model supports noninvasive monitoring of lymphoma progression, immune-cell interactions, and responses to immunotherapies, while reducing the need for repeated tissue collection. In immunology and infection research, it can also help evaluate how host defenses or experimental treatments alter the survival and localization of malignant B cells.

Pressure Transducer: Calibration Using a Pitot-static Tube

2023

Source: Shreyas Narsipur, Mechanical and Aerospace Engineering, North Carolina State University, Raleigh, NC Fluid pressure is an important flow characteristic that is required to determine the aerodynamics of a system. One of the oldest and still existing pressure measurement systems is the manometer due to its accuracy and simplicity of operation. The manometer is generally a U-shaped glass tube that is partially filled with liquid, as shown in Figure 1. The U-tube manometer requires no...

Source: Chang, X., et al. Preparation of Pseudo-Typed H5 Avian Influenza Viruses with Calcium Phosphate Transfection Method and Measurement of Antibody Neutralizing Activity. J. Vis. Exp. (2021).This video demonstrates a luciferase-based assay to quantify pseudovirus infection in epithelial cells. The pseudovirus encodes the luciferase gene, which is expressed in infected cells. After washing, and cells are incubated with lysis buffer at an ultra-low temperature, and then thawed to induce lysis.

Transfection into the macrophage cell line, RAW264.7, is difficult due to the cell’s natural response against foreign materials. We described here a gentle yet robust procedure for transfecting luciferase reporter genes into RAW264.7 cells.

(91), e51890, doi:10.3791/51890 (2014).This video demonstrates the preparation of cell culture samples to assess viral load using a luciferase-based assay. It outlines the process of infecting epithelial cells with a genetically modified virus, transferring viral supernatants and standards into assay wells, and promoting viral entry through centrifugation and incubation. The...
