Peptide Encapsulation

Peptide encapsulation is the incorporation of peptide molecules within protective carriers or materials to improve their stability, delivery, or controlled release. Encapsulation can shield peptides from enzymatic degradation and environmental conditions by enclosing them in systems such as lipid-based particles, polymeric nanoparticles, or biodegradable matrices, which release their cargo through diffusion, degradation, or changes in local conditions. In immunology and infection research, this strategy can enhance antigen presentation, support vaccine development, and deliver antimicrobial or immune-modulating peptides to target tissues. It also enables more consistent dosing and may improve peptide activity in biological environments.

Cellular Encapsulation in 3D Hydrogels for Tissue Engineering

We present protocols for the 3-dimensional (3D) encapsulation of cells within synthetic hydrogels. The encapsulation procedure is outlined for two commonly used methods of crosslinking (michael-type addition and light-initiated free radical mechanisms), as well as a number of techniques for assessing encapsulated cell behavior.

Tracking Hypoxic Signaling within Encapsulated Cell Aggregates

A method for photo-encapsulation of cells in a crosslinked PEG hydrogel is described. Hypoxic signaling within encapsulated murine insulinoma (MIN6) aggregates is tracked using a fluorescent marker system. This system allows serial examination of cells within a hydrogel scaffold and correlation of hypoxic signaling with changes in cell phenotype.
