Viral Mutation Effects

Viral mutation effects are the biological consequences of genetic changes that arise in viral genomes and influence how viruses replicate, spread, and interact with hosts. Mutations commonly result from errors during genome replication, while recombination and natural selection can preserve variants that alter viral proteins, antigenicity, tissue tropism, or resistance to antiviral treatment. In immunology and infection research, analyzing these effects helps explain immune escape, changes in disease severity, and differences in transmission. Genomic surveillance and laboratory studies use this knowledge to track emerging variants, evaluate vaccine effectiveness, guide therapeutic development, and improve outbreak preparedness.

A mutation is a change in the sequence of bases of DNA or RNA in a genome. Some mutations occur during replication of the genome due to errors made by the polymerase enzymes that replicate DNA or RNA. Unlike DNA polymerase, RNA polymerase is prone to errors because it is not capable of “proofreading” its work. We provide a method for testing BRCA1 variants in a tissue culture based assay for homologous recombination repair of DNA damage by depleting endogenous BRCA1 protein from a cell using RNAi and replacing it with a BRCA1 point mutant that contains a coding change.
