Embryonic Mouse Vasculature

Embryonic mouse vasculature is the developing network of blood vessels that delivers oxygen and nutrients, removes waste, and establishes the circulatory environment needed for organ formation. It arises through vasculogenesis, in which endothelial progenitors form initial vessels, and angiogenesis, in which existing vessels sprout, branch, remodel, and mature in response to molecular signals and blood flow. Studying this system reveals how vascular development shapes immune-cell trafficking, tissue inflammation, and host responses to infection. Imaging and genetic analysis of embryonic vessels also help identify mechanisms underlying congenital vascular defects and provide experimental models for evaluating pathways that regulate immunity and vascular integrity.

Developmental studies in the mouse are hampered by the inaccessibility of the embryo during gestation. To promote the long-term culture of the embryonic heart at late stages of gestation, we developed a protocol in which the excised heart is cultured in a semi-solid, dilute Matrigel.
