Bone Decalcification

Bone decalcification is the process of removing mineral salts, primarily calcium, from bone or other calcified tissue so that specimens can be sectioned and examined. It works by exposing tissue to acidic solutions that dissolve calcium compounds or to chelating agents such as EDTA that bind calcium ions, while preserving the collagen-rich organic matrix as much as possible. In medicine, decalcification supports histological analysis, allowing researchers and clinicians to evaluate bone structure, marrow, tumors, infections, and disease-related changes under a microscope. Careful control of the method and duration helps maintain tissue morphology and staining quality.

Immunocytochemical identification of peripheral sensory nerve fiber subtypes (and detection of protein expression therein) are key to the understanding of molecular mechanisms underlying peripheral sensation. Here we describe methods for preparation of peripheral/visceral tissue samples, such as skin and limb bones, for specific immunostaining of peripheral sensory nerve fibers.

Murine Hind Limb Long Bone Dissection and Bone Marrow Isolation

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
