Modified Scruff Grip

Modified Scruff Grip is a laboratory animal-handling technique used to restrain mice safely and consistently during biomedical procedures. The handler gently grasps the loose skin over the shoulders and neck while supporting the animal’s body and hindquarters, limiting movement without compressing the airway or restricting breathing. In medicine and laboratory research, this approach can facilitate physical examination, injections, dosing, blood collection, and other brief interventions that require controlled positioning. Proper training, calm handling, and appropriate support help reduce the risk of escape, struggling, and handling-related injury, improving both animal welfare and the reliability of experimental procedures.

2023

Source: Laboratories of Margaret Workman and Kimberly Frye - Depaul University Genetic modification of foods has been a controversial issue due to debated concerns over health and environmental safety. This experiment demonstrates technical understanding of how food DNA is genetically identified, allowing for educated decision making about the safety and potential dangers of using genetically modified organisms (GMOs) in food supplies. Polymerase Chain Reaction (PCR) is used to amplify food DNA...

Modified Boxplots

A standard box and whisker plot informs us about the spread of the data in a given sample. One can identify the minimum value, maximum value, first quartile value, second quartile or median value, and third quartile. However, the box plot does not tell the reader about outliers - values that lie far from the center of the data. We can modify the standard box and whisker plot to identify the outliers and visualize the actual spread of the data in a sample. Initially, we calculate the adjusted...

Transfecting Primary Macrophages with Modified mRNA

This video demonstrates the process of transfecting primary macrophages with modified mRNA encoding green fluorescent protein. These modified mRNAs, designed to reduce immunogenicity and improve its stability, ensure the successful transfection and subsequent expression of green fluorescent proteins, which is confirmed through fluorescence microscopy.

A Phenotyping Regimen for Genetically Modified Mice Used to Study Genes Implicated in Human Diseases of Aging

2016

A reverse-genetics approach to understanding gene families associated with human disease is presented, using mouse as a model system, and the subsequent mouse phenotyping schedule is described. Because mice defective in a gene of interest, HtrA2, manifested Parkinsonian symptoms, the phenotyping regimen is focused on identifying neurological defects.
