Murine Dissection

Murine dissection is the systematic opening and examination of a mouse to identify organs, tissues, and anatomical relationships, making it a foundational technique in biomedical education and research. Following approved animal-care and biosafety procedures, the specimen is positioned, the skin and body wall are carefully incised, and tissues are reflected or removed in sequence to preserve structures for observation, sampling, or analysis. This approach supports anatomical training, pathology assessment, developmental studies, and evaluation of disease models or treatment effects. Careful dissection also links gross anatomy with histology, physiology, and experimental findings, helping researchers interpret how structural changes relate to medical outcomes.

Pathophysiological changes in the cardiac autonomic nervous system, especially in its sympathetic branch, contribute to the onset and maintenance of ventricular arrhythmias. In the present protocol, we show how to characterize murine stellate ganglia to improve the understanding of the underlying molecular and cellular processes.

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.

A Dissection Method to Harvest Murine Popliteal Draining Lymph Nodes for Ex Vivo Imaging

This video demonstrates a dissection technique to harvest popliteal draining lymph nodes from the mouse hind legs. Lymph nodes play a critical role in bridging innate and adaptive immunity, and harvesting intact lymph nodes — with a preserved tissue structure and cellular viability — can help study local and systemic immune responses.

Here we present a protocol for in vitro isolation of multiple glial cell populations from a mouse CNS. This method allows for the segregation of regional microglia, oligodendrocyte precursor cells, and astrocytes to study the phenotypes of each in a variety of culture systems.
