Murine Heart Injection

Murine heart injection is a preclinical technique for delivering cells, drugs, genetic vectors, or tracing materials directly into a mouse heart, enabling controlled study of cardiac biology and disease. Under anesthesia, a fine needle is positioned in a cardiac chamber or myocardial tissue, where the injected material distributes locally or enters the circulation; injection route, volume, and placement influence delivery and tissue response. Researchers use this method to model myocardial injury, evaluate regenerative or gene-based therapies, track cardiac cell behavior, and assess treatment safety. Its reproducibility and physiological relevance make it valuable in cardiovascular medicine and translational research.

Intramyocardial cell delivery in murine models of cardiovascular diseases, such as hypertension or myocardial infarction, is widely used to test the therapeutic potential of different cell types in regenerative studies. Therefore, a detailed description and a clear visualization of this surgical procedure will help to define the limits and advantages of cardiovascular cell therapeutic analyses in small rodents.

Supramolecular hydrogelators based on ureido-pyrimidinones allow full control over the macroscopic gel properties and the sol–gel switching behavior using pH. Here, we present a protocol for formulating and injecting such a supramolecular hydrogelator via a catheter delivery system for local delivery directly in relevant areas in the pig heart.
