Platelet Quality Control

Platelet quality control is the systematic testing of platelet concentrates to confirm their safety, integrity, and ability to support hemostasis before transfusion. It combines measurements such as platelet count, volume, pH, morphology or swirling, and selected functional or sterility assessments, while monitoring storage conditions and changes over time. In transfusion medicine, these checks help detect collection, processing, or storage problems, support compliance with blood-component standards, and reduce the risk of ineffective transfusion. Consistent quality control also generates performance data that guide product handling, storage practices, and improvements in platelet manufacturing and patient care.

We describe a straightforward method for the isolation of washed platelets from human blood followed by agonist-induced platelet aggregation measurements by turbidimetry. As an example we apply this method for studying the aggregation response of human platelets to collagen after a pre-incubation with the Pannexin1 channel inhibitor Brilliant Blue FCF.

Purification and Activation of Platelets from Murine Whole Blood

In this video, we describe a protocol for purifying platelets from murine whole blood and their subsequent activation with the platelet agonist thrombin. When analyzed with flow cytometry, the purified resting platelets are characterized based on the expression of their cell-surface marker CD41, whereas purified activated platelets co-express both the surface markers CD41 and CD62P.
