Brain Injection Technique

Brain injection technique is a precision method for delivering substances directly into defined regions of the brain, enabling researchers to study neural structure and function. Guided by stereotaxic coordinates, a needle or cannula is positioned through a small cranial opening, and controlled pressure deposits a drug, tracer, viral vector, or other material within brain tissue. In neuroscience, this approach supports investigations of neural circuits, neurochemical signaling, gene expression, and disease mechanisms. Careful targeting and control of injection volume help limit tissue disruption and improve experimental consistency, making the technique valuable for behavioral studies, circuit manipulation, therapeutic research, and validation of experimental models.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.

MicroRNAs play crucial roles in the brain and are potential targets for modeling neuro-degeneration. However, perturbing miRNA levels is challenging due to the short length of miRNA and inaccessibility of the brain tissue. This video presents a method for antagomir design and brain specific delivery using a neuropeptide in mice.

Intracarotid Cancer Cell Injection to Produce Mouse Models of Brain Metastasis

Brain metastasis has become an urgent unmet medical need as its incidence has increased while therapeutic options have remained palliative. Creating experimental animal models of brain metastasis via intracarotid arterial injection of cancer cells facilitates mechanistic studies of the disease biology and evaluation of novel intervention regimens.

A Hypertonic Saline Injection Technique for Inducing Glaucoma in Rats

This video demonstrates glaucoma induction in an anesthetized rat by injecting hypertonic saline into the episcleral vein. The saline enters the trabecular meshwork via Schlemm's canal, causing tissue scarring, which blocks aqueous humor outflow and raises intraocular pressure. The elevated pressure damages retinal ganglion cells, leading to glaucoma.

Stereotactic injection of lentiviruses expressing cDNAs or shRNAs can modulate gene expression in specific brain areas of mice. Here, we present a protocol to combine stereotactic injections with behavioral tasks, such as the Open Field Test (OFT) and the Forced Swim Test (FST).
