Camp Release Measurement

cAMP release measurement is the quantification of cyclic adenosine monophosphate exported from cells, providing a way to study how this second messenger communicates beyond its site of synthesis. In a typical assay, neuronal or glial cells are stimulated, extracellular cAMP is collected or monitored, and its concentration is determined while adenylyl cyclase production and phosphodiesterase-mediated degradation shape the signal. In neuroscience, these measurements help characterize neuromodulatory signaling, neuron–glia communication, and changes in cellular responsiveness. They can also support studies of receptor pathways, pharmacological regulation, and disease-related disruptions in intracellular and extracellular messenger dynamics.

Characterizing the function of odorant receptors serves an indispensable part in the deorphanization process. We describe a method to measure the activation of odorant receptors in real time using a cAMP assay.

Single Cell Measurement of Dopamine Release with Simultaneous Voltage-clamp and Amperometry

The amperometric technique measures dopamine release from a single cell by detecting the oxidative current produced by spontaneous dopamine oxidization. Simultaneous voltage clamp and amperometry methodology reveal the mechanistic relationship between the overall "activity" of dopamine transporter and the regulatory role of this activity on the reverse transport of dopamine.

Exosomes are microvesicular structures found within biofluids that potentially carry important disease discriminatory biomarkers. Here, a novel method is used to specifically extract exosomes and rapidly test the exosomal cargo for both RNA/protein targets following the disruption of exosomes using non-uniform electric cyclic square waves.

The protocol presented in this study illustrates the effectiveness of the cAMP Difference Detector In Situ in measuring cAMP through two methods. One method utilizes a 96-well plate reading spectrophotometer with HEK-293 cells. The other method demonstrates individual HASM cells under a fluorescent microscope.

Measuring Mast Cell Exocytosis via Neuropeptide Y Monomeric Red Fluorescent Protein Release

2025

This video demonstrates mast cell exocytosis induced by an activating reagent that includes a calcium ionophore, initiating the release of secretory granules containing the fluorescent reporter protein NPY-mRFP. Subsequently, we quantitatively assess the exocytosis by measuring the released NPY-mRFP using a fluorescence plate reader.
