Cell Type Specific Expression

Cell type-specific expression is a strategy for restricting a gene or reporter to a defined population of cells, enabling researchers to distinguish cellular functions within complex tissues. In neuroscience, this selectivity is achieved by placing genes under cell-type-specific promoters or enhancers, or by combining targeted viral or transgenic constructs with recombinase systems such as Cre-lox. The approach supports selective labeling, calcium imaging, optogenetic control, and genetic manipulation of neurons or glial cells. By linking gene activity to cellular identity, cell type-specific expression helps map neural circuits, clarify circuit function, and improve the precision of disease models and potential therapies.

Cell Specific Gene Expression

2020

Multicellular organisms contain a variety of structurally and functionally distinct cell types, but the DNA in all the cells originated from the same parent cells. The differences in the cells can be attributed to the differential gene expression. Liver cells, whose functions include detoxification of blood, production of bile to metabolize fats, and synthesis of proteins essential for metabolism, must express a specific set of genes to perform their functions. Gene expression also varies with...

The molecular basis of spatial-specific phytochrome responses is being investigated using transgenic plants that exhibit tissue- and organ-specific phytochrome deficiencies. The isolation of specific cells exhibiting induced phytochrome chromophore depletion by Fluorescence-Activated Cell Sorting followed by microarray analyses is being utilized to identify genes involved in spatial-specific phytochrome responses.

Translating ribosome affinity purification (TRAP) enables rapid and efficient isolation of cell type-specific translating mRNA. Here, we demonstrate a method that combines hydrodynamic tail-vein injection in a mouse model of liver repopulation and TRAP to examine the expression profile of repopulating hepatocytes.
