Hippocampal Glutamate

Hippocampal glutamate is the principal excitatory neurotransmitter system in the hippocampus, where it supports communication between neurons involved in learning and memory. Glutamate released from presynaptic terminals activates AMPA and NMDA receptors on neighboring cells; receptor activity depolarizes the postsynaptic membrane, permits calcium influx through NMDA receptors, and can produce long-term synaptic strengthening. Glutamate transporters and the glutamate–glutamine cycle help regulate signaling and limit excessive stimulation. Studying hippocampal glutamate clarifies the cellular basis of synaptic plasticity and informs research on memory disorders, epilepsy, excitotoxicity, and other conditions involving disrupted neural communication.

An In Vitro Technique to Study Glutamate Receptor Trafficking in Hippocampal Neurons

This video demonstrates an antibody-feeding approach to study the trafficking of glutamate receptors in primary hippocampal neuronal cultures. This approach allows the observation of receptor populations at both the plasma and internal membranes. Discrimination between these subsets is achieved by labeling the receptors before and after membrane permeabilization, using the same primary antibody but secondary antibodies conjugated to different fluorophores.

This article presents a method to study glutamate receptor (GluR) trafficking in dissociated primary hippocampal cultures. Using an antibody-feeding approach to label endogenous or overexpressed receptors in combination with pharmacological approaches, this method allows for the identification of molecular mechanisms regulating GluR surface expression by modulating internalization or recycling processes.

Glutamate and Hypoxia as a Stress Model for the Isolated Perfused Vertebrate Retina

With this study, we introduce a standardized stress model for the isolated superfused bovine retina for future preclinical therapeutic testing. The effect of either hypoxia (pure N2) or glutamate stress (250 µM glutamate) on retinal function represented by a- and b-wave amplitudes was evaluated.

Organotypic Hippocampal Slice Cultures

We describe a method to prepare organotypic hippocampal slices that can be easily adapted to other brain regions. Brain slices are laid on porous membranes and culture media is allowed to form an interface. This method preserves the gross architecture of the hippocampus for up to 2 weeks in culture.
