In Vitro Preparation

In vitro preparation is the isolation and maintenance of cells, tissues, or organs outside an organism under controlled laboratory conditions, allowing biological functions to be studied directly. In neuroscience, researchers preserve neural tissue or culture neurons in nutrient-containing media or artificial extracellular solutions, while controlling factors such as temperature, oxygenation, ionic composition, and stimulation to support viability and measurable activity. These preparations enable precise investigation of synaptic transmission, neuronal excitability, circuit function, and responses to drugs or injury. By separating neural mechanisms from the complexity of a living organism, in vitro approaches support electrophysiology, imaging, disease modeling, and the development of experimental and therapeutic strategies.

Preparation and Analysis of In Vitro Three Dimensional Breast Carcinoma Surrogates

We demonstrate a method to generate 3D breast cancer surrogates, which can be cultured using a perfusion bioreactor system to deliver oxygen and nutrients. Following growth, surrogates are fixed and processed to paraffin for evaluation of parameters of interest. The evaluation of one such parameter, cell density, is explained.

Implant-associated infection is a significant clinical complication. This study describes an approach using platelet-rich plasma (PRP) to prevent implant-associated infections, presents the protocol for preparing PRP with constant platelet concentration, and reports the newly identified antimicrobial properties of PRP and related protocols for examining such antimicrobial properties in vitro.

Preparation and In Vitro Characterization of Magnetized miR-modified Endothelial Cells

This manuscript describes the efficient, non-viral delivery of miR to endothelial cells by a PEI/MNP vector and their magnetization. Thus, in addition to genetic modification, this approach allows for magnetic cell guidance and MRI detectability. The technique can be used to improve the characteristics of therapeutic cell products.

Here, we present the preparation of an er-xian decoction (EXD) in four steps—soaking, decoction, filtration, and concentration—and demonstrate the administration of a prepared EXD-containing serum to rats. These methods are applicable to the in vivo and in vitro study of herbal decoctions such as traditional Chinese medicines.
