Olfactory Behavior Assay

Olfactory behavior assays are experimental methods that measure how organisms detect, discriminate, and respond to odors, providing a behavioral readout of sensory and neural function. During an assay, controlled odorants are presented under defined conditions, while researchers record outcomes such as approach, avoidance, investigation, preference, habituation, or odor-guided learning. These behaviors arise as olfactory receptors convert chemical cues into neural signals that engage brain circuits involved in perception, motivation, memory, and decision-making. In neuroscience, olfactory behavior assays help evaluate sensory processing, study neural circuit function, and identify changes associated with development, aging, neurological disease, or genetic manipulation.

We describe here a behavioral setup and data analysis method for assaying olfactory responses of up to 100 vinegar flies (Drosophila melanogaster). This system may be used with single or multiple olfactory stimuli, and adaptable for optogenetic activation or silencing of neuronal subsets.

This video demonstrates a study of the navigational behavior of Drosophila larvae in response to optogenetic stimulation of olfactory sensory neurons (ORNs). Transgenic Drosophila larvae expressing light-sensitive ion channels in the ORNs are placed on agarose within a behavior arena. Optogenetic stimulation with light opens the channels, causing a cation influx that triggers the transmission of the signal to the brain, and the navigation behavior is recorded.
