Rectal Suction Biopsy

Rectal suction biopsy is a minimally invasive diagnostic technique that removes small samples of rectal mucosa and submucosa to evaluate the enteric nervous system, especially when Hirschsprung disease is suspected. A suction device captures tissue without requiring a full-thickness surgical incision, allowing histological examination for the presence, absence, or abnormal distribution of enteric ganglion cells and supporting nerve fibers. In neuroscience and pediatric gastrointestinal research, the procedure helps connect intestinal motility disorders with defects in neural development and signaling. Its findings can guide diagnosis, surgical planning, and investigation of how enteric neurons regulate coordinated bowel function.

Immunohistochemical Staining of S-100 Protein in Human Rectal Suction Biopsy Sections

This video demonstrates the immunohistochemical staining for S-100 protein in rectal suction biopsy sections. Antigen-retrieved sections are blocked and treated with primary, followed by secondary antibodies against S-100, a marker of Hirschsprung's disease. Chromogenic substrate is added to stain the targeted cells. Samples are dehydrated and mounted before visualizing under the microscope.
