Slice Culture Preparation

Slice culture preparation is a neuroscience technique that maintains thin, living sections of brain or spinal cord tissue outside the organism for controlled study. Tissue is rapidly dissected, sectioned with a vibratome or related instrument, and placed in nutrient-rich culture medium under conditions that support oxygenation, cellular survival, and synaptic function. These preparations preserve local neural circuits while providing access for microscopy, electrophysiology, pharmacological manipulation, and genetic or viral labeling. Researchers use slice cultures to investigate neuronal connectivity, synaptic plasticity, neurodevelopment, and mechanisms of neurological disease, linking cellular responses to the organization of intact neural tissue.

The chicken auditory brainstem is comprised of nuclei responsible for binaural sound processing. A single coronal slice preparation maintains the entire circuitry while the cultured approach provides a unique preparation to study the development of neuronal structure and auditory function at the molecular, cellular and network levels.

We describe the preparation of thymic slices that, in combination with flow cytometry, can be used to model positive and negative selection of developing T cells. Thymic slices can also be adapted for the in situ analysis of thymocyte migration, localization, and signaling via immunofluorescence and two-photon microscopy.

Preparation of Organotypic Slice Cultures from a Rat Pup's Hippocampus

2025

This video demonstrates a method for preparing organotypic hippocampal slice cultures from the brain of a rat pup. It outlines the steps involved in the dissection of the hippocampus, slicing, and culture conditions required to promote cell viability and growth in the hippocampal slices, ultimately generating organotypic slice cultures.

Preparing Free-Floating Slice Cultures from an Adult Human Brain
