Slice Recovery Procedure

The slice recovery procedure is a preparation step that allows freshly cut brain tissue slices to regain physiological stability before neuroscience experiments. After sectioning, slices are placed in oxygenated artificial cerebrospinal fluid, often under controlled temperature and timing conditions, so damaged cells can re-establish ionic balance, restore metabolic function, and recover network responsiveness. This process improves the quality and reproducibility of acute-slice recordings, calcium imaging, and pharmacological studies by supporting more viable tissue and consistent neuronal activity. Careful recovery conditions are therefore essential for investigating synaptic transmission, circuit function, and cellular responses in ex vivo brain preparations.

2025

This video describes a protocol to obtain brain slices using an N-Methyl-D-glucamine (NMDG)-based protective recovery method. A dissected brain is incubated in an aCSF with NMDG and HEPES to minimize neuronal cell death and damage during the sectioning. The section is allowed to recover in the aCSF to restore the cellular...

Lung-tumor Tissue Explant Culture: A Procedure to Generate and Culture Precision-cut Lung Tumor Slices

2023

Organotypic primary tissue explant cultures, which include precision-cut slices, represent the three-dimensional (3-D) tissue architecture as well as the multicellular interactions of native tissue. This video describes the culturing of murine lung tumor-derived tissue slices using a rotating incubation unit, a system that enables intermittent exposure of tissues to oxygen and nutrients. The protocol describes the methodology to generate and culture precision-cut lung tumor slices for drug...

Obtaining Horizontal Mouse Hippocampal Brain Slices

2025

This video demonstrates a detailed protocol for preparing horizontal hippocampal brain slices from mice, focusing on preserving the integrity of hippocampal fiber pathways. The process involves carefully bisecting and securing the brain, using a vibratome for slicing, and maintaining optimal tissue conditions with a carbogenated fluid, resulting in hippocampal brain slices ready for further experimentation.
