Working with Centrifuges

1. Algemeen protocol voor veilig centrifuge-gebruik
<ol>
	<li>De laboratoriumsupervisor of veiligheidsofficier moet ervoor zorgen dat alle gebruikers goed zijn getraind in de selectie van de rotor, monsterbuis en de juiste werking van de centrifuge.</li>
	<li>Elke nieuwe gebruiker moet nauwgezet worden begeleid door een getrainde gebruiker totdat hij/zij zeker is van hoe het instrument correct te gebruiken.</li>
	<li>Voorafgaand aan het gebruik van het instrument, moet elke gebruiker de veiligheidsinstructies voor de specifieke centrifuge zorgvuldig lezen en begrijpen.</li>
	<li>Afhankelijk van het monster, moeten compatibele rotoren worden gebruikt.</li>
	<li>De rotor, buis en spil moeten schoon, droog en vrij van scheuren of misvormingen zijn.</li>
	<li>De O-ringen moeten worden gecontroleerd op slijtage of scheuren.</li>
	<li>Het is belangrijk om ervoor te zorgen dat de buizen die worden gebruikt voor centrifugatie compatibel zijn met het monster en de rotatiesnelheid (laag, hoog of ultra).</li>
	<li>De centrifugebuizen mogen niet meer dan driekwart van hun maximale vulniveau worden gevuld.</li>
	<li>De buizen moeten goed worden afgesloten.</li>
	<li>Het is uiterst belangrijk om de buizen tegenover elkaar te balanceren. Als dit niet gebeurt, kan dit leiden tot onvolledige scheiding en in extreme gevallen kan dit leiden tot verspreiding van de inhoud van de buis in het behuizing van de centrifuge.</li>
	<li>Voorafgaand aan het inschakelen van de centrifuge, moet de rotor worden gecontroleerd om er zeker van te zijn dat deze correct op de aandrijving zit en dat de deksel goed gesloten is.</li>
	<li>De deksel moet altijd gesloten worden gehouden wanneer de centrifuge in gebruik is, en mag nooit worden geopend totdat de rotor volledig is gestopt.</li>
	<li>De operator moet in de nabijheid van de centrifuge blijven totdat deze de doelsnelheid heeft bereikt. In het geval van ongebruikelijke geluiden of trillingen, moet het instrument onmiddellijk worden uitgeschakeld. De snelheidslimiet van het instrument mag nooit worden overschreden. Laat bij het uitschakelen voldoende tijd verstrijken om de rotor tot stilstand te brengen. Moderne centrifuges hebben indicatoren die aangeven dat het veilig is om de centrifuge te openen.</li>
	<li>Nadat de centrifugatie voorbij is, moeten de monsters worden verwijderd en moet het instrument worden gecontroleerd op eventuele lekkages of vloeistoffen. Indien nodig, moet de rotor grondig worden gereinigd.</li>
</ol>
2. Aanvullende voorzorgsmaatregelen bij het hanteren van gevaarlijke chemicaliën of besmettelijke materialen
<ol>
	<li>Bij het gebruik van gevaarlijke chemicaliën of biologische monsters die mogelijk besmettelijk zijn, is het belangrijk om het instrument tijdens het gebruik te labelen om andere werknemers op de hoogte te stellen van het gevaar.</li>
	<li>Er moeten geschikte handschoenen1 worden gedragen om gevaarlijke monsters te hanteren.</li>
	<li>Er moeten afgesloten veiligheidsbuizen worden gebruikt. Het is wenselijk om secundaire insluiting te gebruiken, indien beschikbaar, voor de centrifugebuizen. Deze zijn meestal in de vorm van plastic of glazen inzetstukken die direct in de monsterbakken van de centrifuge worden geplaatst.</li>
	<li>Nadat de centrifugatie voorbij is, moeten de buizen uit de rotor worden verwijderd en alleen worden geopend in de afzuigkap of biologische veiligheidskast, indien van toepassing.</li>
	<li>Reiniging, indien nodig, moet met uiterste zorg worden uitgevoerd, om de operator niet bloot te stellen aan gevaarlijke chemicaliën of besmettelijke materialen. In geval van lekkage van dergelijke monsters, zie stap 3, Noodsituaties, hieronder.</li>
	<li>Voor specifieke soorten gevaarlijke monsters kan het beter zijn om standaard procedures en reinigingsprotocollen in het laboratorium te handhaven. Deze moeten worden beoordeeld en bijgewerkt, indien nodig.</li>
</ol>
3. Noodsituaties en hun veiligheidsmaatregelen
Een noodsituatie kan zich voordoen als de rotor defect raakt of als er een lekkage in de centrifuge optreedt als gevolg van breuk van een buis of onjuist gebruik. In dergelijke situaties moeten de volgende stappen worden ondernomen.
<ol>
	<li>De centrifuge moet onmiddellijk worden uitgeschakeld en de deksel moet gesloten worden gehouden totdat anderen op de hoogte zijn gebracht en de locatie hebben verlaten. De stroom moet worden onderbroken via de aan/uit-knop op het instrument. De knop bevindt zich op de voorkant, zijkant of achterkant van het instrument. De gebruiker moet zich vertrouwd maken met de locatie. Als de aan/uit-knop defect is, moet de gebruiker de hoofdstekker uit het stopcontact verwijderen.</li>
	<li>Om het risico op aerosolen te verminderen, is het soms beter om de centrifuge minimaal 0,5 uur onaangeroerd te laten. Raadpleeg de technische documentatie van de centrifuge.</li>
	<li>Voor het opruimen van lekkages moet de operator goed bekend zijn met het gebruikte monster en moet hij/zij de juiste kleding dragen, inclusief een laboratoriumjas, handschoenen, gezichtsbescherming en veiligheidsbril.
	<ol>
		<li>Na reiniging moet het gelekte materiaal op de juiste manier worden afgevoerd en moet de besmette beschermende kleding worden afgevoerd of op de juiste manier worden schoongemaakt. Handen en alle blootgestelde huid moeten grondig worden gewassen.</li>
	</ol>
	</li>
	<li>Indien overduidelijk blootgesteld aan zeer gevaarlijke materialen, moet 911 worden gebeld of moet onmiddellijk medische hulp worden verleend aan de getroffen persoon.</li>
</ol>
4. Onderhoud van de centrifuge
OPMERKING: De behuizing van de centrifuge is meestal gemaakt van metalen die kunnen corroderen in aanwezigheid van vocht, chemicaliën of sterke reinigingsmiddelen. Voor goed onderhoud van de centrifuge is het aan te raden om de instructies van de fabrikant voor reiniging en onderhoud zorgvuldig op te volgen. Behalve dat, zijn er enkele algemene maatregelen voor onderhoud waarmee rekening moet worden gehouden, zoals hieronder.

	<li>De centrifuge moet altijd schoon en droog worden gehouden.</li>
	<li>Elke vorm van lekkage

Met het toegenomen gebruik van centrifuges in laboratoria en de industrie, is ook het risico op gevaren bij het gebruik van centrifuges toegenomen. Echter, zorgvuldige omgang met het apparaat en goede kennis van het gebruik en veiligheidsmaatregelen kunnen ongevallen aanzienlijk verminderen. De zwaartekracht van de aarde is in staat om heterogene mengsels te scheiden. Veel systemen vermijden echter dit type scheiding vanwege de tijd die in dergelijke gevallen meestal vereist is. Centrifugatie is een van de krachtigste hulpmiddelen voor het scheiden van heterogene mengsels1. Het omvat de toepassing van middelpuntvliedende kracht voor sedimentatie van de zwaardere fase, wat leidt tot scheiding van de twee fasen, en is een veelgebruikte techniek in industriële en laboratoriumomgevingen.
Bij een typisch centrifugatieproces worden deeltjes die in een vloeistof zijn opgeschort, rond de rotatieas van de centrifuge gedraaid, waardoor een kracht van de as wordt veroorzaakt. Deze kracht drijft de deeltjes naar de bodem van de centrifuge-monsterhouder met een snelheid die afhankelijk is van de grootte en dichtheid van de deeltjes. Centrifugatie is dus een techniek om het sedimentatieproces te versnellen.
Om de veiligheid bij het gebruik van een centrifuge te waarborgen, moet de gebruiker goed getraind zijn en zich bewust zijn van de gevaren die kunnen voortvloeien uit het gebruik ervan. De volgende secties behandelen de afhandeling en veiligheid bij het werken met centrifuges.

Centrifugeren is een van de krachtigste hulpmiddelen die in laboratoria worden gebruikt voor het scheiden van heterogene mengsels, maar het kan gevaarlijk zijn en daarom moeten veiligheidsmaatregelen worden toegepast.?Een centrifuge draait snel monsters om heterogene materialen te scheiden, wat extreme centrifugale krachten op de monsters en het instrument veroorzaakt. Vanwege de hoge snelheid waarmee een centrifuge werkt, kan het ernstig letsel bij de gebruiker veroorzaken als het verkeerd wordt gehanteerd. Daarom moet er speciale zorg worden genomen bij het gebruik van een centrifuge om morsen en schade aan het apparaat te voorkomen.?Deze video zal het mechanisme van een centrifuge illustreren en hoe je er veilig mee kunt werken en het onderhoud ervan kunt uitvoeren.Voordat we ingaan op de details van de veiligheidsmaatregelen, laten we snel de basisprincipes van centrifugatie bekijken. Centrifuges bevatten een rotor die op een ashuis is geplaatst. Op een enkele as kunnen meestal rotors van verschillende grootte worden gemonteerd om verschillende monsterflessen, buizen of zakken te bergen. Een motor draait de rotor met een bepaald aantal RPM of omwentelingen per minuut. Dit brengt op zijn beurt de centrifugale kracht over op het heterogene mengsel, gericht weg van de rotatieas, wat deeltjes met verschillende dichtheden scheidt.Verschillende soorten centrifuges werken op verschillende RPM's. Lage-snelheidscentrifuges draaien met minder dan 10.000 RPM, hoge-snelheid tot 21.000 en ultracentrifuges tot 30.000 en hoger. Elk type heeft verschillende rotors die ermee compatibel zijn. Na de draai wordt de rotor meestal afgeremd. In elke centrifuge vervormt hoge snelheid de rotor.Bij normaal gebruik zal het terugkeren naar zijn vorige grootte. Als de kracht van het draaien echter de maximale kracht die de rotor aankan overschrijdt, kan deze permanent vervormd raken. Dit kan leiden tot scheuren, wat op zijn beurt tot ernstige storingen kan leiden. Centrifuge-storingen kunnen aan de andere kant leiden tot het lekken van monsters, wat gevaarlijk kan zijn als er gevaarlijke materialen bij betrokken zijn en dient voorzichtig te worden behandeld. Nu laten we u zien hoe u monsters voorbereidt en veilig gebruikt in de centrifuge.Selecteer eerst geschikte centrifugebuizen die compatibel zijn met de rotatiesnelheden die u wilt gebruiken. Bekijk de documentatie om de limieten van de buis te achterhalen. Buizen moeten intact zijn en vrij van scheuren. Vul ze tot maximaal 3/4 om morsen te voorkomen. Zorg er vervolgens voor dat de buizen uitgebalanceerd zijn en sluit ze goed af.Neem speciale voorzorgsmaatregelen bij het centrifugeren van gevaarlijke chemische of biologische monsters. Draag geschikte beschermende uitrusting en bereid monsters voor in een geschikte werkomgeving, zoals een afzuigkap of laminaire stromingskast. Zie de video's van JoVE over afzuigkappen voor meer informatie over deze veiligheidsfuncties.Gebruik indien nodig een secundaire beveiliging voor gevaarlijke materialen, zoals een glazen of plastic inzetstuk. Verzegelde veiligheidsbuizen die een gesloten omgeving bieden, kunnen ook worden gebruikt. Iedereen moet worden getraind voordat hij een centrifuge of rotor gebruikt.Selecteer een rotor die geschikt is voor de centrifuge. Controleer of de rotor en de afdekking schoon en droog zijn en geen scheuren of misvormingen hebben. Zorg er extra voor dat de O-ringen op de rotor of afdekking niet versleten zijn. Controleer ook de centrifuge-as om te zien of deze gebogen is of andere misvormingen heeft. Plaats vervolgens de rotor op de as en zorg ervoor dat deze correct zit. Plaats monsters in de rotor en zorg ervoor dat de buizen goed in de rotor passen.Plaats altijd uitgebalanceerde buizen tegenover elkaar. Bedek de rotor en zorg ervoor dat de afdichting goed is. Sluit het deksel van de centrifuge en start uw programma. Zie nogmaals de vorige centrifuge-video van JoVE voor meer informatie over het uitvoeren van een centrifugeprogramma.Blijf bij de centrifuge totdat deze zijn maximale snelheid heeft bereikt. Maak uzelf vertrouwd met de locatie van de aan/uit-knop. Schakel de stroom uit als er ongebruikelijke geluiden of trillingen optreden om de centrifuge onmiddellijk te stoppen. Evacueer vervolgens het directe gebied totdat de rotor is gestopt. Zodra deze is gestopt, controleert u de monsters op lekkage en de rotor op eventuele afwijkingen of onbalans. Als er geen problemen zijn geweest, wacht u tot de centrifuge zijn programma heeft voltooid en volledig is gestopt.Wacht nog een half uur als er aerosolvormende chemicaliën aanwezig zijn. Anders mag ude centrifuge onmiddellijk openen. Verwijder de monsters en breng ze terug naar een gespecialiseerd gebied voor opening indien nodig. In geval van morsen, reinig de centrifuge onmiddellijk. Gebruik geschikte PPE voor elk aanwezig gevaar en verwijder verontreinigd materiaal volgens de richtlijnen. Gebruik geen schurende borsteldraden om te reinigen, omdat deze de rotor kunnen beschadigen. Waarschuwt noodpersoneel en probeer de centrifuge niet schoon te maken als er levensbedreigende gevaren zijn gemorst.?Decontaminatie van de rotor met 10% bleekmiddel voor alle biologische monsters die in de centrifuge zijn gebruikt, ongeacht of er is gemorst of niet. Laat het bleekmiddel 30 minuten staan en spoel vervolgens met 70% ethanol en laat het drogen. Verwijder en inspecteer de rotor ten slotte op eventuele schade en noteer uw gebruik in het logboek.Je hebt zojuist JoVE's Introductie in centrifuge-gebruik bekeken.
