Phase Contrast and Differential Interference Contrast Microscopy

Unstained living cells are transparent and hard to see under an optical microscope. Phase-contrast and differential-interference contrast or DIC microscopes allow their visualization by creating higher resolution and sharper contrast images.
When a sample is illuminated in-phase-contrast, some light is diffracted by the cell components, while the rest directly passes through. The extent of diffraction depends on the refractive indices of the cell components.
As the diffracted and the direct light interfere, parts of the sample appear bright if the waves are in-phase and dark if they are out-of-phase. This creates high-contrast images of large organelles and thin tissue sections.
Sharp changes in refractive indices produce a halo around the image.
Such artifacts are absent in DIC, which uses a polarized light source to modulate the light waves passing through the sample.
The image contrast depends on the rate of change of the refractive indices across the specimen. Steep changes generate high-contrast images with an apparent three-dimensional effect.
DIC helps visualize thick specimens like brain slices, eggs, or embryos.

Fasecontrastmicroscopen

Met in-fasecontrastmicroscopen wordt interferentie tussen licht dat rechtstreeks door een cel gaat en licht dat door cellulaire componenten wordt gebroken, gebruikt om contrastrijke beelden met hoge resolutie te creëren zonder vlekken. Het is het oudste en eenvoudigste type microscoop dat een beeld creëert door de golflengten van de lichtstralen die door het preparaat gaan te veranderen. Veranderde golflengtepaden worden gecreëerd met behulp van een ringvormige stop in de condensor. De ringvormige stop produceert een holle lichtkegel die op het preparaat wordt gefocust voordat deze de objectieflens bereikt. Het objectief bevat een faseplaat met een fasering. Als gevolg hiervan gaat licht dat rechtstreeks van het belichtingstoestel komt door de fasering, terwijl licht dat door het monster wordt gebroken of gereflecteerd door de plaat gaat. Dit zorgt ervoor dat golven die door de ring reizen ongeveer de helft van een golflengte uit fase zijn met de golven die door de plaat gaan.

Omdat golven pieken en dalen hebben, kunnen ze optellen (als ze in fase zijn) of elkaar opheffen (als ze uit fase zijn). Wanneer de golflengten uit fase zijn, heffen golfdalen de golfpieken op, wat destructieve interferentie wordt genoemd. Structuren die licht breken, zien er vervolgens donker uit tegen een heldere achtergrond van alleen niet-gebroken licht. Meer in het algemeen zullen structuren die verschillen in kenmerken zoals de brekingsindex, verschillen in de mate van duisternis. Omdat het het contrast vergroot zonder dat er vlekken nodig zijn, wordt fasecontrastmicroscopie vaak gebruikt om levende exemplaren te observeren. Specifieke cellulaire structuren, zoals organellen in eukaryotische cellen en endosporen in prokaryotische cellen, worden goed zichtbaar gemaakt met fasecontrastmicroscopie.

Differentiële interferentiecontrastmicroscopen

Differentiële interferentiecontrastmicroscopen (DIC) (ook bekend als Nomarski-optica) zijn vergelijkbaar met fasecontrastmicroscopen in die zin dat ze interferentiepatronen gebruiken om het contrast tussen verschillende kenmerken van een monster te verbeteren. In een DIC-microscoop ontstaan twee lichtbundels waarbij de richting van de golfbeweging (polarisatie) verschilt. Zodra de bundels door het monster of de monstervrije ruimte gaan, worden ze opnieuw gecombineerd. De effecten van de monsters veroorzaken verschillen in de interferentiepatronen die worden gegenereerd door het combineren van de bundels. Dit resulteert in contrastrijke beelden van levende organismen met een driedimensionaal uiterlijk. Phase-contrast and differential interference contrast microscopy help scientists see live cells without stains. Both methods boost contrast so thin, transparent structures become easier to study under the microscope.

Phase-contrast microscopes use interference between light that passes straight through a cell and light that is refracted by cell parts. The microscope uses an annular stop in the condenser to make a hollow cone of light. The objective lens also contains a phase plate with a phase ring. Light from the illuminator passes through the ring, while refracted light passes through the plate. This makes the light waves about one-half wavelength out of phase.

When waves are out of phase, their peaks and troughs can cancel each other out. This is called destructive interference. As a result, structures that bend light appear dark against a bright background. Differences in refractive index can also change how dark a structure looks. Because it does not require stains, phase-contrast microscopy is often used for live specimens. It can clearly show organelles in eukaryotic cells and endospores in prokaryotic cells.

Differential interference contrast (DIC) microscopy, also called Nomarski optics, uses a similar idea. It creates two beams of light with different polarization directions. After the beams pass through the specimen or through empty space, they are recombined. The specimen changes the interference pattern that forms when the beams come together.

DIC microscopy produces high-contrast images of living organisms with a three-dimensional look. It is especially useful for telling apart structures inside live, unstained specimens. Like phase-contrast microscopy, it helps reveal details that are hard to see with ordinary bright-field light.
