8-oxo-dg Quantification

8-oxo-dG quantification is the measurement of 8-oxo-2′-deoxyguanosine, a major oxidatively modified DNA nucleoside and a biomarker of oxidative damage. In a typical workflow, DNA is isolated and enzymatically hydrolyzed to nucleosides, then 8-oxo-dG is separated and measured using high-performance liquid chromatography with electrochemical detection, liquid chromatography-mass spectrometry, or immunoassays against calibrated standards. Accurate analysis requires minimizing artificial oxidation during sample preparation and often normalizing 8-oxo-dG to total deoxyguanosine. In biochemistry, these measurements help assess reactive oxygen species, DNA repair activity, cellular stress, disease-associated damage, and the effectiveness of antioxidant or protective interventions.
