Cyclodextrin Mediated Exchange

Cyclodextrin-mediated exchange is a biochemical method that transfers lipids, especially cholesterol, between biological membranes or membrane models using cyclodextrin carriers. Cyclodextrins are cyclic oligosaccharides with a hydrophobic cavity that temporarily encapsulates lipid molecules while their hydrophilic exterior supports movement through aqueous solutions; differences in lipid concentration drive exchange between donor and acceptor membranes. Researchers use this approach to selectively alter membrane composition, investigate cholesterol distribution and lipid asymmetry, and study how membrane properties influence protein activity and cell signaling. The method provides controlled manipulation of membrane lipids for biochemical and biophysical experiments.

A quick and efficient method to integrate foreign DNA of interest into pre-made acceptor strains, termed landing pad strains, is described. The method allows site-specific integration of a DNA cassette into the engineered landing pad locus of a given strain, through conjugation and expression of the ΦC31 integrase.

The motion regimes of ibuprofen encapsulated in β-cyclodextrin nanosponges polymer network are investigated using pulsed-field-gradient spin-echo (PGSE) NMR technique. Synthesis, purification, drug loading, implementation of the NMR pulse sequence and data analysis to work out the mean square displacement of the drug at several observation times are described in detail.

Proteins often contain multiple domains that can exert different cellular functions. Gene knock-outs (KO) do not consider this functional diversity. Here, we report a recombination-mediated cassette exchange (RMCE)-based structure-function approach in KO embryonic stem cells that allows for the molecular dissection of various functional domains or variants of a protein.

Rapid and Efficient Generation of Recombinant Human Pluripotent Stem Cells by Recombinase-mediated Cassette Exchange in the AAVS1 Locus

Here we report a rapid and efficient gene editing method based on RMCE in the AAVS1 locus of human Pluripotent Stem Cells (hPSCs) that improves upon previously described systems. Using this technique, isogenic lines can be rapidly and reliably generated for proper comparative studies, facilitating transgenesis-mediated research with hPSCs.
