Large Scale Protein Production

Large-scale protein production is the controlled manufacture of recombinant or native proteins in quantities sufficient for research, diagnostics, industrial processes, or therapeutic development. In biochemistry, the process typically introduces a protein-encoding gene into a suitable host such as bacteria, yeast, or cultured mammalian cells, then grows the cells in bioreactors under conditions that support gene expression, protein folding, and biomass formation. The target protein is subsequently harvested from cells or culture media and purified using techniques selected for its physical and chemical properties. Scaled production provides consistent material for structural studies, enzyme characterization, drug screening, vaccine development, and commercial bioprocessing, while presenting challenges in yield, quality, stability, and downstream purification.

Synthetic Spider Silk Production on a Laboratory Scale

Despite the outstanding mechanical and biochemical properties of spider silks, this material cannot be harvested in large quantities by conventional means. Here we describe an efficient strategy to spin artificial spider silk fibers, which is an important process for investigators studying spider silk production and their use as next-generation biomaterials.

MultiBac is a baculovirus/insect cell system particularly tailored for expressing eukaryotic proteins and their complexes. MultiBac was implemented as an open-access platform, and standard operating procedures developed to maximize its utility.
