2d And 3d Culture

2D and 3D culture are laboratory systems for growing cells in controlled environments, providing complementary models for studying biology and engineering living tissues. In 2D culture, cells attach to and spread across a flat substrate, while 3D culture organizes cells within aggregates, scaffolds, or biomaterial matrices that better reproduce cell-cell interactions, extracellular matrix contact, and chemical gradients. Bioengineers use these models to investigate cell behavior, disease processes, drug responses, and tissue formation. Comparing 2D and 3D systems helps researchers select appropriate experimental platforms and design more physiologically relevant constructs for regenerative medicine and tissue engineering.

We present a protocol to obtain cell-derived matrices rich in extracellular matrix proteins, using macromolecular crowders (MMC). In addition, we present a protocol which incorporates MMC in 3D organotypic skin co-culture generation, which reduces culture time while maintaining maturity of construct.

Chromosome painting is a useful method for studying organization of the cell nucleus and evolution of the karyotype. Here, we demonstrate an approach to isolate and amplify specific regions of interest from single polytene chromosomes that are subsequently used for two- and three-dimensional fluorescent in situ hybridization (FISH).

This video describes the technique for long term 2D co-culture of leukemia cells with bone marrow stromal cells to study their interaction with the bone marrow microenvironment. The 2D coculture model utilizes the acute lymphoblastic leukemia cells and the bone marrow stromal or osteoblasts that influence tumor cell phenotype. This study will support investigation of the mechanisms that underlie bone marrow supported survival of leukemic cells during chemotherapy exposure.
