Bone Mechanobiology

Bone mechanobiology is the study of how mechanical forces regulate bone structure, function, and adaptation, making it central to both skeletal biology and bioengineering. During loading, osteocytes sense tissue strain and fluid movement within the bone matrix, then transmit signals that influence osteoblast-mediated formation and osteoclast-mediated resorption through coordinated remodeling. This framework helps explain how bones maintain strength, respond to exercise or injury, and lose mass during disuse. In bioengineering, bone mechanobiology guides the design of implants, biomaterial scaffolds, and mechanical stimulation strategies that promote integration, regeneration, and durable skeletal repair.

Murine Hind Limb Long Bone Dissection and Bone Marrow Isolation

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.

This article describes the experimental procedures used to prepare rat tail tendons for biomechanical and mechanobiological studies. Several features of the main steps in preparation are demonstrated, beginning with extraction, cross-sectional area measurement, rinsing and loading into the bioreactor chamber.

In the example, we perform bone marrow aspiration from an acute myeloid leukemia (AML) patient and examine the cells to check for measurable residual disease (MRD), an important prognostic biomarker.
