High-throughput Cytometry

High-throughput cytometry is a rapid, automated approach for measuring physical and molecular features across large numbers of cells or particles, enabling quantitative analysis of heterogeneous biological systems. In flow-based instruments, cells are suspended in fluid, aligned for optical interrogation, and passed through laser beams; detectors record light scattering and fluorescence from labeled targets, while automated sample handling increases measurement capacity. In bioengineering, the method supports cell population profiling, biomarker analysis, screening of engineered cells, and evaluation of biomaterials or bioprocess conditions. Its scale and multiparametric data help researchers identify rare populations, optimize designs, and assess biological performance efficiently.

Over the recent years, live cell-based assays have been used successfully to detect antibodies against surface and conformational antigens. Here, we describe a method using high-throughput flow cytometry enabling the analysis of large cohorts of patients. Detection of novel antibodies will improve diagnosis and treatment of immune-mediated disorders.

A protocol for nanoparticle tracking analysis (NTA) and high-throughput flow cytometry to evaluate polymeric gene delivery nanoparticles is described. NTA is utilized to characterize the nanoparticle particle size distribution and the plasmid per particle distribution. High-throughput flow cytometry enables quantitative transfection efficacy evaluation for a library of gene delivery biomaterials.

Macropinocytosis, large-scale non-specific fluid uptake, is important in many areas of clinical biology including immunology, infection, cancer, and neurodegenerative diseases. Here, existing techniques have been adapted to allow high-throughput, single-cell resolution measurement of macropinocytosis in the macropinocytosis model organism Dictyostelium discoideum using flow cytometry.
