Spasticity Assessment

Spasticity assessment is the systematic evaluation of abnormal, velocity-dependent increases in muscle tone and reflex activity, providing information about neuromuscular impairment and movement limitations. During testing, an examiner or robotic device applies a controlled passive stretch while recording resistance, joint angle, movement velocity, and, in instrumented systems, muscle activity through electromyography. Clinical scales offer practical bedside ratings, whereas biomechanical measurements can quantify torque and distinguish neural contributions from passive tissue stiffness. In bioengineering, these data support rehabilitation planning, assistive-device design, treatment monitoring, and development of more objective models of motor dysfunction.

Children with spastic cerebral palsy (SCP) have limb spasticity, movement disorders, and abnormal posture due to injury to the cerebral cortex motor area, resulting in inability to stand and walk normally. Therefore, alleviating limb spasticity and enhancing gross motor function in children with SCP have become important therapeutic goals.

Impaired mitochondrial transport and morphology are involved in various neurodegenerative diseases. The presented protocol uses induced pluripotent stem cell-derived forebrain neurons to assess mitochondrial transport and morphology in hereditary spastic paraplegia. This protocol allows characterization of mitochondrial trafficking along axons and analysis of their morphology, which will facilitate the study of neurodegenerative disease.

Isokinetic Robotic Device to Improve Test-Retest and Inter-Rater Reliability for Stretch Reflex Measurements in Stroke Patients with Spasticity

Using a robotic isokinetic device with electromyography (EMG) measurements, this protocol illustrates that isokinetic motion itself can improve inter-rater reliability for the angle of catch measurements in stroke patients with mild elbow flexor spasticity.
