Vesicle Encapsulation

Vesicle encapsulation is the process of enclosing therapeutic, diagnostic, or biological cargo within membrane-bound compartments, enabling controlled protection, transport, and release. In bioengineering, amphiphilic lipids or polymers self-assemble in aqueous environments to form vesicles with an internal aqueous core and, in some systems, a hydrophobic membrane that partitions cargo according to its chemical properties; preparation conditions influence vesicle size, stability, and encapsulation efficiency. This approach supports drug delivery, protection of proteins and nucleic acids from degradation, and localized release at target tissues, while providing a platform for engineering material properties and improving the performance of biomedical therapies.

Giant vesicles containing highly packed micrometer-sized components are useful cell models. The water-in-oil emulsion centrifugation method is a simple, powerful tool for the preparation of giant vesicles with encapsulated materials.

Herein we describe simple methods for the preparation of vesicles, the encapsulation of transcription and translation machinery, and the monitoring of protein production. The resulting cell-free systems can be used as a starting point from which to build increasingly complex cellular mimics.

Rapid Encapsulation of Reconstituted Cytoskeleton Inside Giant Unilamellar Vesicles

This article introduces a simple method for expeditious production of giant unilamellar vesicles with encapsulated cytoskeletal proteins. The method proves to be useful for bottom-up reconstitution of cytoskeletal structures in confinement and cytoskeleton-membrane interactions.

Tracking Hypoxic Signaling within Encapsulated Cell Aggregates

A method for photo-encapsulation of cells in a crosslinked PEG hydrogel is described. Hypoxic signaling within encapsulated murine insulinoma (MIN6) aggregates is tracked using a fluorescent marker system. This system allows serial examination of cells within a hydrogel scaffold and correlation of hypoxic signaling with changes in cell phenotype.
