Astrocyte Culture

Astrocyte culture is an in vitro method for isolating, growing, and maintaining astrocytes, the glial cells that support neuronal function and help regulate the brain’s extracellular environment. Typically, neural tissue is dissociated mechanically or enzymatically, and cells are expanded on treated culture surfaces in defined or serum-containing media under controlled temperature, gas, and nutrient conditions; enrichment may rely on differences in adhesion and growth. These cultures model astrocyte morphology, proliferation, inflammatory signaling, neurotransmitter handling, and interactions with neurons, while enabling controlled testing of neurotoxic compounds and disease mechanisms. Although they do not reproduce the full architecture of brain tissue, astrocyte cultures complement animal models in neurobiology and pharmacology research.

Astrocytes have been recognized to be versatile cells participating in fundamental biological processes that are essential for normal brain development and function, and central nervous system repair. Here we present a rapid procedure to obtain pure mouse astrocyte cultures to study the biology of this major class of central nervous system cells.

Generating a Primary Culture of Astrocytes from a Rat Pup Brain

The video demonstrates a technique for culturing primary astrocytes from the rat cortex. The cortex is mechanically dissociated to obtain a homogeneous glial cell suspension, which is then plated on polypeptide-coated plates to promote selective proliferation of astrocytes. Subsequently, the astrocytes are enzymatically dissociated from the plates and transferred to fresh media, allowing the culture of pure astrocytes.

Upon establishing the co-culture of motor neurons and astrocytes on a multi-electrode array (MEA) plate, the synchronous rhythmic electrical activity of the neuronal network was recorded extracellularly using the MEA.
