Gnotobiotic Conditions

Gnotobiotic conditions are controlled biological environments in which researchers know and regulate the microorganisms associated with an organism, making them essential for studying host–microbe interactions. Animals or other experimental systems are maintained in sterile isolators and either kept germ-free or deliberately colonized with defined microbial communities under carefully monitored conditions. These models allow investigators to separate the effects of individual microorganisms from those of complex microbiota and to examine how microbial composition influences development, immunity, metabolism, and disease. In biology, gnotobiotic research supports mechanistic microbiome studies, testing of microbial therapies, and more reproducible experimental outcomes.

Rearing the Fruit Fly Drosophila melanogaster Under Axenic and Gnotobiotic Conditions

2016

A method for rearing Drosophila melanogaster under axenic and gnotobiotic conditions is presented. Fly embryos are dechorionated in sodium hypochlorite, transferred aseptically to sterile diet, and reared in closed containers. Inoculating diet and embryos with bacteria leads to gnotobiotic associations, and bacterial presence is confirmed by plating whole-body Drosophila homogenates.

This protocol is used in establishing and maintaining gnotobiotic American cockroaches (Periplaneta americana) by surface sterilizing the egg cases (oothecae) prior to hatching. These gnotobiotic insects contain their vertically transmitted Blattabacterium endosymbionts but have axenic guts.

A Gnotobiotic System for Studying Microbiome Assembly in the Phyllosphere and in Vegetable Fermentation

A method of growing germ-free Napa cabbages has been developed which enables researchers to evaluate how single microbial species or multispecies microbial communities interact on cabbage leaf surfaces. A sterile vegetable extract is also presented which can be used to measure shifts in community composition during vegetable fermentation.

2026

This protocol describes the assembly and use of a new plant gnotobiotic growth system designed to accommodate cereals and validated in wheat and maize. This plant gnotobiotic system enables controlled plant–microbe experiments by providing sterile growth conditions, irrigation management, and compatibility with diverse soil-based substrates.

Fear Conditioning

2023

Fear Conditioning is a type of learning in which an association is established between a negative unpleasant event and a harmless stimulus. This leads to a fear of the harmless stimulus. This process is largely mediated by the amygdala, which is a brain region involved in emotions and stress reactions. Fear conditioning can be utilized in several ways to understand different aspects of learning and memory. This video presents an overview of the principles behind fear conditioning, discusses the...
