Metaphase Spread Analysis

Metaphase spread analysis is a cytogenetic technique used to visualize and evaluate condensed chromosomes, making it possible to assess chromosome number, structure, and arrangement. During the procedure, dividing cells are arrested in metaphase, when chromosomes are most compact, then exposed to hypotonic treatment, fixed, and spread onto microscope slides for staining and imaging. Researchers use the resulting chromosome spreads to construct karyotypes and identify aneuploidy, translocations, deletions, duplications, and other genomic abnormalities. In biology and medical research, this analysis supports studies of cell division, development, cancer, inherited disorders, and chromosome stability.

Oogenesis in mammals is known to be error-prone, particularly due to chromosome missegregation. This manuscript describes chromatin spread preparation methods for mouse prophase, metaphase I and II-staged oocytes. These fundamental techniques allow for the study of chromatin-bound proteins and chromosome morphology throughout mammalian oogenesis.

DNA-FISH Metaphase Spreads to Distinguish Extrachromosomal DNA from Homogeneously Staining Regions in Human Cancer Cell Lines

2026

We report a DNA-FISH metaphase spread protocol that visually detects locus copy number and location within the genome. This approach enables single-cell resolution of amplification states, specifically in cancer cell lines containing extrachromosomal DNA and homogeneously staining regions.

Spreading of Chromatin Modifications

2020

The histone proteins in the nucleosomes are post-translationally modified (PTM) to increase or decrease access to DNA. The commonly observed PTMs are methylation, acetylation, phosphorylation, and ubiquitination of lysine amino acids in the histone H3 tail region. These histone modifications have specific meaning for the cell. Hence, they are called "histone code". The protein complex involved in histone modification is termed as "reader-writer" complex. Writers The writer is an enzyme that can...
