Nematode Culturing

Nematode culturing is the controlled maintenance and propagation of nematode populations under laboratory conditions, enabling researchers to study these microscopic roundworms and their interactions with hosts, microbes, or the environment. Cultures typically provide a suitable growth medium, moisture, temperature, and food source, while careful transfer and contamination control support reproduction and stable population growth. In biology, cultured nematodes serve as experimental models for genetics, development, behavior, ecology, and host-parasite relationships. Standardized culture conditions improve reproducibility and allow researchers to examine life cycles, measure responses to environmental or chemical changes, and test biological hypotheses.

Visualizing Bacteria in Nematodes using Fluorescent Microscopy

To study the mutualism between Xenorhabdus bacteria and Steinernema nematodes, methods were developed to monitor bacterial presence and location within nematodes. The experimental approach, which can be applied to other systems, entails engineering bacteria to express the green fluorescent protein and visualizing, using fluorescence microscopy bacteria within the transparent nematode.

Entomopathogenic nematodes live in symbiosis with bacteria and together they successfully infect insects by undermining their innate immune system. To promote research on the genetic basis of nematode infection, methods for maintaining and genetically manipulating entomopathogenic nematodes are described.

Behavioral assays provide powerful tools for understanding neuronal function. Here we present several protocols for quantifying predatory feeding behavior found in the model nematode Pristionchus pacificus and its relatives. Additionally, we provide methods for analyzing predatory feeding adaptations including mouth structures and teeth.

This video demonstrates an in vivo assay to study mitophagy in Caenorhabditis elegans. Upon exposing the nematodes to a mitophagy-inducing drug, the occurrence of mitophagy is assessed by co-localization between DCT-1, a mitophagy receptor on the damaged mitochondria, and the autophagosomal membrane protein LGG-1.
