Subcutaneous Implantation

Subcutaneous implantation is a biological and experimental procedure in which a device, biomaterial, tissue sample, or therapeutic construct is placed beneath the skin to maintain contact with living tissue. The implanted material interacts with the surrounding extracellular environment, while diffusion, tissue integration, and the local inflammatory or foreign-body response influence its performance and persistence. In biology, researchers use subcutaneous implantation to evaluate biomaterial compatibility, monitor tissue responses, deliver drugs, or study implanted cells and engineered tissues in vivo. Results can reveal how implants function within the body and guide the development of safer medical devices, sustained therapies, and regenerative strategies.

This protocol describes a unique experimental model of implant-related infections that enables the simultaneous incubation of two implants with bacteria under identical conditions within a single mouse. It also allows precise assessment of biofilm formation on implant surfaces using optimized comparative analytical methods, demonstrating advanced techniques for evaluating biomaterials' antimicrobial properties.

We describe the manufacture of compressed hormone pellets, as well as subcutaneous surgical implantation into mice. This strategy can be combined with the growth of cell and tissue xenografts under the renal capsule of athymic nude mice to evaluate hormonal carcinogenesis and regulation of benign prostate growth.

Treating chronic or neuropathic facial pain can be challenging when medical or standard treatment fails. Subcutaneous nerve field stimulation is the least invasive form of neuromodulation and is used for chronic back pain. We applied this technology to treat chronic and neuropathic trigeminal facial pain.

2013

Human tumor xenografts in immunodeficient mice are valuable tools to study cancer biology. Specific protocols to generate subcutaneous and intrahepatic xenografts from human hepatocellular carcinoma cells or tumor fragments are described. Liver regeneration induced by partial hepatectomy in recipient mice is presented as a strategy to facilitate intrahepatic engraftment.
