Zygote Germination

Zygote germination is the transition in which a fertilized cell, or a zygote-derived dormant structure, resumes growth and develops into an embryo or new organism. When suitable conditions activate metabolism, the zygote uses stored nutrients, re-enters the cell cycle, undergoes mitotic division, and differentiates into specialized tissues. This process links sexual reproduction with early development and may depend on environmental cues such as water, temperature, oxygen, and nutrient availability. Studying zygote germination helps researchers understand embryogenesis, developmental timing, reproductive success, and life-cycle transitions across plants, fungi, algae, and other organisms.

To purify zygotes of S. cerevisiae, haploid cells of opposite mating type were engineered to express red or green fluorescent proteins, co-incubated to allow zygote formation, and fractionated using a flow cytometry-based protocol. The highly-enriched fraction enables subsequent "-omic" studies, recovery of initial progeny, and systematic investigation of zygote morphogenesis.

The ability of embryonic germinal cells to differentiate into primordial germinal cells during early development stages is a perfect model to address our hypothesis about cancer and infertility. This protocol shows how to isolate primordial germinal cells from developing gonads in 10.5-11.5 days post coitum mouse embryos.
