3d Culture Protocol

A 3D culture protocol is a method for growing cells within or around a three-dimensional matrix or scaffold, creating an experimental environment that more closely reflects tissue architecture than conventional two-dimensional culture. By supporting cell-cell and cell-matrix interactions, 3D systems promote spatial organization and generate conditions such as nutrient, oxygen, and signaling gradients that influence proliferation, differentiation, and survival. In cancer research, these models can form tumor spheroids, organoids, or matrix-embedded cultures for studying tumor growth, invasion, microenvironmental responses, and treatment effects. Their ability to capture clinically relevant features makes them valuable for mechanistic studies, drug screening, and personalized cancer research.

A 3D system of culturing human articular chondrocytes in high levels of synovial fluid is described. Synovial fluid reflects the most natural microenvironment for articular cartilage, and can be easily obtained and stored. This system thus can be used for studying cartilage regeneration and for screening therapeutics for treating arthritis.
