Bispecific Antibodies

Bispecific antibodies are engineered proteins designed to bind two different molecular targets, creating therapeutic links that conventional monospecific antibodies cannot. In cancer research, one binding site can recognize an antigen on a tumor cell while the other binds CD3 on a T cell; this proximity forms an immune synapse that activates T-cell signaling and promotes targeted cytotoxicity. These molecules support strategies for redirecting immune responses against malignant cells and help investigators study antigen selection, immune activation, and treatment resistance. Their modular design enables combinations of tumor targets and immune effectors, making bispecific antibodies an important platform for developing selective cancer therapies and evaluating how tumors evade immune attack.

An In Vivo Technique to Track Bispecific Antibody-Induced T Cell Trafficking to Tumors

This video demonstrates a method for tracking bispecific antibody-induced T cell trafficking to tumors in a mouse model. Luciferase-transduced effector T cells are injected into a mouse with a subcutaneous tumor cell xenograft. Bispecific antibodies are then introduced, binding to T cells and the tumor cells. The localization of the T cells is tracked using bioluminescent imaging, with the magnitude of the signal indicating the extent of T cell infiltration into the tumor.
