Cell Speed Analysis

Cell speed analysis is a quantitative method for measuring how quickly individual cells or cell populations move, providing insight into migration, invasion, and changes in cellular behavior. It typically uses time-lapse microscopy to record cell positions over defined intervals, followed by tracking and calculation of displacement, distance traveled, or velocity. In cancer research, these measurements help characterize tumor-cell motility, compare invasive potential, and assess responses to genetic changes, drugs, or differences in the surrounding microenvironment. By converting dynamic movement into measurable data, cell speed analysis supports studies of metastasis and improves the evaluation of anti-invasive treatments.

Investigating Outer Hair Cell Motility with a Combination of External Alternating Electrical Field Stimulation and High-speed Image Analysis

A reliable method to investigate outer hair cell (OHC) motile responses, including electromotility, slow motility and bending, is described. OHC motility is elicited by stimulation with an external alternating electrical field, and the method takes advantage of high-speed image recording, LED-based illumination, and last generation image analysis software.

High Speed Sub-GHz Spectrometer for Brillouin Scattering Analysis

Here we present a protocol to build a rapid Brillouin spectrometer. Cascading virtually imaged phase array (VIPA) etalons achieve a measurement speed more than 1,000 times faster than traditional scanning Fabry-Perot spectrometers. This improvement provides the means for Brillouin analysis of tissue and biomaterials at low power levels in vivo.

High-speed Continuous-wave Stimulated Brillouin Scattering Spectrometer for Material Analysis

We describe the construction of a rapid continuous-wave-stimulated-Brillouin-scattering (CW-SBS) spectrometer. The spectrometer employs single-frequency diode-lasers and an atomic vapor notch-filter to acquire transmission spectra of turbid/non-turbid samples with high spectral-resolution at speeds up to 100-fold faster than those of existing CW-SBS spectrometers. This improvement enables high-speed Brillouin material analysis.

Here, we present a protocol to isolate the plasma membrane, cytoplasm and mitochondria of U937 cells without the use of high-speed centrifugation. This technique can be used to purify subcellular fractions for subsequent examination of protein localization via immunoblotting.

High-speed video microscopy analysis is a relatively easy-to-perform, fast, cost-effective, and, in experienced hands, a considerably reliable tool for first-line diagnostics of primary ciliary dyskinesia, which should be available in every center involved in diagnostics and the treatment of severe lung diseases.
