Acid Induced Assembly

Acid-induced assembly is a chemical process in which an acid stimulus drives molecules, polymers, or nanoscale building blocks to organize into larger, ordered structures. The acid changes the protonation state of reactive groups, altering electrostatic interactions, hydrogen bonding, solubility, or molecular shape and thereby shifting the balance toward aggregation or network formation. Researchers use this stimulus-responsive behavior to control supramolecular structures, gels, capsules, and other functional materials under defined chemical conditions. Because assembly can be triggered or reversed by adjusting acidity, the approach supports studies of molecular recognition, sensing, drug delivery, catalysis, and the design of adaptive materials.

This protocol describes a clinically-applicable means of dissolving hydrophobic compounds in an aqueous environment using combinations of self-assembling peptide and amino acid solutions. Our method resolves a major limitation of hydrophobic therapeutics, which lack safe, efficient means of solubility and delivery methods into clinical settings.

The analytical ultracentrifuge (AUC) sample cell holds sample and reference buffer and during experiments and is exposed to high vacuum and rotor speeds up to 60,000 rpm. This video will demonstrate the rigorous attention to detail necessary for assembly, loading and alignment of this very important component of an AUC experiment.

Protein Complex Assembly

2020

Proteins can form homomeric complexes with another unit of the same protein or heteromeric complexes with different types. Most protein complexes self-assemble spontaneously via ordered pathways, while some proteins need assembly factors that guide their proper assembly. Despite the crowded intracellular environment, proteins usually interact with their correct partners and form functional complexes. Many viruses self-assemble into a fully functional unit using the infected host cell to...
