Brain Ventricular Injection

Brain ventricular injection is a method for delivering cells, drugs, nucleic acids, or other substances directly into the fluid-filled ventricles of the brain, making it valuable for studying neural development. A fine needle or micropipette introduces the material into a cerebral ventricle, where cerebrospinal fluid can distribute it across nearby brain regions and developing neural tissues. In developmental biology, researchers use this approach to manipulate gene expression, alter signaling pathways, trace cell behavior, or test therapeutic compounds during embryonic and postnatal development. The technique provides targeted experimental control and helps reveal how molecular cues regulate neurogenesis, cell migration, differentiation, and tissue organization.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.

The injected virus circulates through the cerebrospinal fluid, infecting cells in the periventricular region, including the marginal zone, choroid plexus, and subventricular zone.
