Cardiac Reprogramming

Cardiac reprogramming is the conversion of differentiated, non-cardiac cells into cardiomyocyte-like cells by altering their gene-regulatory state, offering a way to study heart development and repair. The process typically uses cardiac transcription factors or other molecular signals to activate cardiomyocyte gene networks while suppressing the original cellular identity through transcriptional and epigenetic remodeling. In developmental biology, this approach helps reveal how cardiac cell fate is established and maintained. It also supports disease modeling, investigation of cellular plasticity, and the development of strategies for myocardial regeneration, although researchers must assess reprogramming efficiency, cell maturity, and functional integration.

We describe here, the establishment and application of an Tg(Myh6-cre)1Jmk/J /Gt(ROSA)26Sortm38(CAG-GCaMP3)Hze/J (referred to as αMHC-Cre/Rosa26A-Flox-Stop-Flox-GCaMP3 below) mouse reporter line for cardiac reprogramming assessment. Neonatal cardiac fibroblasts (NCFs) isolated from the mouse strain are converted into induced cardiomyocytes (iCMs), allowing for convenient and efficient evaluation of reprogramming efficiency and functional maturation of iCMs via calcium (Ca2+) flux.

This video showcases the direct reprogramming of astrocytes expressing transforming factors into neurons. Transformation factors trigger a cascade of molecular events facilitating astrocyte-to-neuron conversion. The process is further supported by supplying specific agents and supplements through a neuronal differentiation medium.

Electrophysiological Recordings of Reprogrammed Neurons in a Mouse Brain Slice

This video demonstrates a protocol to evaluate the maturation and reprogramming of glial cells into neurons in a transfected mouse striatum slice. The experiment measures resting membrane potential and response to current injections using neuron-specific fluorescent markers and electrophysiological techniques. Voltage spikes during these measurements indicate successful neuronal maturation and reprogramming.

The protocol presented in this study describes methods for the real-time monitoring of reprogramming progression via the kinetic measurement of positive and negative pluripotent stem cell markers using flow cytometry analysis. The protocol also includes the imaging-based assessment of morphology, and marker or reporter expression during iPSC generation.
