Drug Permeability Testing

Drug permeability testing is a method for measuring how readily a drug crosses a biological barrier, an important step in predicting absorption, distribution, and tissue exposure. In controlled assays, researchers place a compound across a cell layer, membrane, or tissue model and quantify its movement over time, often under defined concentration and temperature conditions. These measurements reveal how molecular size, polarity, transport proteins, and barrier properties influence passage. In developmental biology, permeability testing helps assess whether experimental compounds can reach developing tissues or embryos, supporting studies of signaling pathways, toxicity, therapeutic effects, and the design of safer drugs.

Testing Epithelial Permeability in Fetal Tissue-Derived Enteroids

This protocol details the establishment of enteroids, a three-dimensional intestinal model, from fetal intestinal tissue. Immunofluorescent imaging of epithelial biomarkers was used for model characterization. Apical exposure of lipopolysaccharides, a bacterial endotoxin, using microinjection technique induced epithelial permeability in a dose-dependent manner measured by the leakage of fluorescent dextran.
