Epidermal Volume Extrapolation

Epidermal volume extrapolation is a quantitative method for estimating the three-dimensional volume of an epidermal tissue from measured dimensions or sampled regions. It typically combines epidermal thickness, surface area, and geometric or spatial assumptions to extend local measurements across a larger tissue or developmental structure. In developmental biology, this approach helps quantify changes in epidermal growth, organization, and tissue expansion when direct volumetric measurement is impractical. Comparing extrapolated volumes across developmental stages can support analysis of morphogenesis and provide standardized measurements for linking tissue-scale changes with underlying cellular processes.

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.

Pharmacologic Induction of Epidermal Melanin and Protection Against Sunburn in a Humanized Mouse Model

Epidermal melanin is induced by topical application of forskolin in a murine model of the fair-skinned UV-sensitive human. Pharmacologic manipulation of cAMP levels in the skin and epidermal darkening strongly protect against UV-mediated inflammation (sunburn) as measured by the minimum erythematous dose (MED) assay.
