Semen Collection

Semen collection is the controlled acquisition of ejaculate containing spermatozoa and seminal fluid for laboratory analysis, reproductive procedures, or research. Typically, semen is obtained through ejaculation into a sterile collection vessel, then handled under controlled conditions to preserve sample integrity and may be evaluated for sperm concentration, motility, morphology, and viability. In developmental biology, collected semen provides material for studying gamete function, fertilization, early embryonic development, and factors that influence reproductive success. Standardized collection and processing support reliable experiments, assisted reproduction, and investigations into how sperm quality affects embryo formation and offspring development.

This paper presents an economical and efficient protocol for examining peroxidase-positive leukocytes in semen. With the assistance of a computer-assisted semen analysis (CASA) system, the concentration of peroxidase-positive leukocytes in semen can be obtained within a total of 60 min, effectively improving the efficiency of andrology laboratory and andrologists.
