Cii Mutant Scoring

Cii mutant scoring is a phenotypic approach for evaluating how genetic mutations alter an organism’s response to infection, helping link genotype with immune function. The method compares Cii mutant individuals with appropriate controls under standardized conditions and assigns scores to predefined infection-associated traits, producing a consistent measure of resistance, susceptibility, or altered immune activity. In immunology and infection research, these comparisons help identify genes that regulate host defense, distinguish components of immune pathways, and prioritize candidates for further mechanistic study. Quantitative scoring also supports reproducible screening across mutant populations and experimental conditions.

We describe a detailed protocol for the Lambda Select cII mutation assay in cultured cells of transgenic rodents or the corresponding animals treated with a chemical/physical agent of interest. This approach has been widely used for mutagenicity testing of carcinogens in mammalian cells.

Paradigms for Pharmacological Characterization of C. elegans Synaptic Transmission Mutants

This video demonstrates how to employ two neural stimulants, aldicarb and pentylenetetrazole (PTZ), in complementary ways to study synaptic function in the nematode, C. elegans. This complementary approach may also be used to shed light on evolutionarily conserved mechanisms for modulating neuronal synchrony and has implications for epilepsy and seizures.

Shifting Zebrafish Lethal Skeletal Mutant Penetrance by Progeny Testing

The goal of this protocol is to alter the penetrance of lethal skeletal mutant phenotypes in zebrafish by selective breeding. Lethal mutants cannot be grown to adulthood and bred themselves, therefore this protocol describes a method for tracking and selecting penetrance through multiple generations by progeny testing.

Introduction to z Scores

2023

A z score (or standardized value) is measured in units of the standard deviation. It tells you how many standard deviations the value x is above (to the right of) or below (to the left of) the mean, μ. Values of x that are larger than the mean have positive z scores, and values of x that are smaller than the mean have negative z scores. If x equals the mean, then x has a zero z score. It is important to note that the mean of the z scores is zero, and the standard deviation is one. z scores help...

z scores are the standardized values obtained after converting a normal distribution into a standard normal distribution. A z score is measured in units of the standard deviation. The z score tells you how many standard deviations the value x is above (to the right of) or below (to the left of) the mean, μ. Values of x that are larger than the mean have positive z scores, and values of x that are smaller than the mean have negative z scores. If x equals the mean, then x has a z score of zero.
