Murine Stromal Cells

Murine stromal cells are nonhematopoietic support cells derived from mouse tissues that organize local tissue environments and regulate interactions among immune, epithelial, and vascular cells. They function through extracellular-matrix production, cell adhesion, and secretion of cytokines and chemokines, which influence leukocyte survival, migration, activation, and tissue repair during homeostasis or inflammation. In immunology and infection research, primary murine stromal cells and stromal cell lines are used in co-culture systems and tissue-mimicking models to examine immune-cell recruitment, inflammatory signaling, pathogen effects on tissue niches, and host responses to treatment. These models help connect cellular mechanisms with disease progression while enabling controlled experimental comparisons.

This protocol describes an easily adaptable migration assay that uses rigorous statistics to quantify changes in MSC migratory capacity after damage.

Isolation of lymph node stromal cells is a multistep procedure including enzymatic digestion and mechanical disaggregation to obtain fibroblastic reticular cells, lymphatic and blood endothelial cells. In the described procedure, a short digestion is combined with automated mechanical disaggregation to minimize surface marker degradation of viable lymph node stromal cells.

Generating Schwann Cell-Like Cells from Bone Marrow Stromal Cells

This video demonstrates a method for obtaining Schwann cell-like cells from multipotent bone marrow stromal cells (BMSCs). Initially, the BMSCs undergo hypoxia treatment, followed by directed differentiation into neuronal progenitor cells. Subsequently, specific media and culture conditions are employed to induce the transformation of these progenitor cells into Schwann cell-like cells.
